The oxidation of glucose by Penicillium chrysogenum.

چکیده
Studies on the mechanism of glucose metabolism by penicillia are few. The fact that Snell (1949) and Kita (1951) found none of the phosphorylated esters of the Embden-Meyerhof scheme in PeniciUium chrysogenum, strain Q176, and that penicillia contain glucose oxidase (Birkinshaw and Raistrick, 1943) would indicate a priori that glucose is metabolized oxidatively by the mold. The following data indicate that gluco is metabolized oxidatively by P. chrysogenum, strain NRRL 1951-B25, although none of the specific intermediary steps has been determined.

Formate as an auxiliary substrate for glucose-limited cultivation of Penicillium chrysogenum: impact on penicillin G production and biomass yield.
Production of beta-lactams by the filamentous fungus Penicillium chrysogenum requires a substantial input of ATP. During glucose-limited growth, this ATP is derived from glucose dissimilation, which reduces the product yield on glucose. The present study has investigated whether penicillin G yields on glucose can be enhanced by cofeeding of an auxiliary substrate that acts as an energy source b...
متن کاملResolving phenylalanine metabolism sheds light on natural synthesis of penicillin G in Penicillium chrysogenum.
The industrial production of penicillin G by Penicillium chrysogenum requires the supplementation of the growth medium with the side chain precursor phenylacetate. The growth of P. chrysogenum with phenylalanine as the sole nitrogen source resulted in the extracellular production of phenylacetate and penicillin G. To analyze this natural pathway for penicillin G production, chemostat cultures w...
متن کاملMetabolic engineering of β-oxidation in Penicillium chrysogenum for improved semi-synthetic cephalosporin biosynthesis.
Industrial production of semi-synthetic cephalosporins by Penicillium chrysogenum requires supplementation of the growth media with the side-chain precursor adipic acid. In glucose-limited chemostat cultures of P. chrysogenum, up to 88% of the consumed adipic acid was not recovered in cephalosporin-related products, but used as an additional carbon and energy source for growth. This low efficie...
متن کاملThe oxidation of tricarboxylic acid cycle compounds by Penicillium chrysogenum.
The presence of a tricarboxylic acid cycle in Penicillium chry8ogenum, as yet, has not been demonstrated conclusively. Ajl (1951) stated that of the mechanisms proposed for oxidation of the 2 carbon fragment in mold metabolism, the evidence for a tricarboxylic acid cycle is the least convincing. This paper presents a study of some of the enzymes of the tricarboxylic acid cycle in cell-free prep...
متن کاملA reporter gene analysis of penicillin biosynthesis gene expression in Penicillium chrysogenum and its regulation by nitrogen and glucose catabolite repression.
Vectors which possess a truncated niaD gene encoding nitrate reductase were developed to allow targeted gene integration during transformation of an niaD mutant Penicillium chrysogenum host. The Penicillium genes pcbC and penAB are immediately adjacent to each other and are divergently transcribed, with an intergenic control region serving as their promoters. Gene fusions were constructed with ...
